With the persona of a washed up Vegas type comedian, Hamburger's live act features a barrage of question/answer jokes aimed often at celebrity targets as well as depressing barbs aimed at his ex-wife. His pacing is off, and he often clears his throat during his routine, usually to keep overzealous fans from shouting out his punchlines. He has performed in front of audiences worldwide from Madison Square Garden to Jimmy Kimmel Live! and the confrontational aspects of his act have drawn cursory comparisons to Tony Clifton.

His first full length album, America's Funnyman, was released in 1996. He has released a number of albums on the Drag City record label.

Hamburger is a frequent guest on the internet talk show Tom Green's House Tonight. In 2006, Hamburger started doing his own show called Poolside Chats with Neil Hamburger on The Channel at Tomgreen.com. Guests on his show have included Tim and Eric, Kyle Gass of Tenacious D, Buzz Osborne of The Melvins, and Bonnie 'Prince' Billy.

In 2007, Hamburger appeared in the music video for "Conspiracy of the Gods" by the band Trans Am.

In September 2007 and 2010, he appeared at the Global ComedyFest in Vancouver, Canada. In 2010, he appeared at Just For Laughs in Montreal, and later that summer, at the Edinburgh Festival Fringe resulting in rave reviews in The Scotsman and The Guardian.

In spring of 2008, Drag City released an album of country and western originals and covers recorded by Neil Hamburger in a "celebrity vocals" style. During the sessions, Hamburger was backed by musicians including Prairie Prince of The Tubes and Todd Rundgren's band, David Gleason, Atom Ellis of Dieselhed and The New Cars, Rachel Haden from that dog.

In December 2010, Special Entertainment released an iPhone App called Shaky Advice from Neil Hamburger that functions much like a Magic 8 Ball, with 30 video clips of Hamburger giving comical advice.

In 2010 Neil Hamburger opened for Faith No More during their reunion tour, with jokes primarily about Mike Patton's rival Anthony Kiedis.

Neil Hamburger is the invention of Gregg Turkington, who, prior to creating the awkward comedian character, was heavily involved in the underground music scene of Northern California as a band member, collaborator, media creator, and manager. He rose to prominence in the mid-1980s as the co-publisher of underground zine Breakfast Without Meat, which brought punk respectability to "square" musical artists such as Tiny Tim, Jimmy Webb, the 101 Strings, and Richard Harris through feature articles. A frequent contributor to the zine was Derrick Bostrom of the Meat Puppets.

In the 1980s, Turkington also performed in obscure San Francisco bands such as Hello Kitty on Ice and Bean Church, and was rumoured to have been a founding member of Caroliner.

In the early 1990s, Turkington operated the Amarillo Records label, which released works by many artists from San Francisco's burgeoning post-punk avant-garde music scene. He was a member of Totem Pole of Losers and the Zip Code Rapists (with John Singer). He collaborated with Trey Spruance of Mr. Bungle in The Three Doctors, Faxed Head, Secret Chiefs 3 and The Bon Larvis Band. Many of these bands appeared on 1996s landmark Amarillo Records compilation CD, You Gan't Boar Like An Eabla When You Work With Turkrys, along with the likes of Sun City Girls, Thinking Fellers Union Local 282, U.S. Saucer, Harvey Sid Fisher, Dieselhed, and Church of Satan founder Anton Szandor LaVey. He was also a tour manager for both Link Wray and Mr. Bungle. He contributed lead vocals to two tracks by the Secret Chiefs 3.
